    Research Activities:

    Research Area:

HIV is the most significant risk factor for many opportunistic infections like fungal, tuberculosis, etc. The intense media coverage of AIDS puts the public on an emotional roller caster, and inflated promises are engendered by each success. For the moment drugs acting as both HIV-reverse transcriptase inhibitors along with antimicrobial properties are one of the brightest hope. My research focuses in drug design of a novel anti-HIV agent with broad spectrum of chemotherapeutic properties that include anti-tubercular, anti-fungal, anti-HCV, and antibacterial properties. The research efforts of Dr. D. Sriram combine synthetic organic chemistry with computational, biochemical, and molecular biological tools in an interdisciplinary approach to designing drugs.

Anti-tuberculosis drug discovery and development

 

Infection with the intracellular pathogens Mycobacterium sp. can lead to death in animals and humans. In the case of tuberculosis, current epidemiological evidence suggests that one-third of the world's population are infected with M. tuberculosis, that 8 million new cases emerge annually and that 3 million deaths per year are directly attributable to infection with the bacillus. New infection, as well as reactivation of latent tuberculosis, is particularly prevalent in individuals with compromised immune systems, such as those that are HIV positive. In addition, the emergence of drug-resistant strains of M. tuberculosis has led to increased pressure on current chemotherapy regimes. We are using both natural products chemistry and rational synthesis as models for the discovery of new anti-tuberculosis agents. To date we have identified several classes of compounds that exhibit useful potency (MIC < 1 μg/mL) towards both wild-type and drug resistant strains of M. tuberculosis. We are currently preparing new analogues in an effort to develop more potent derivatives with improved pharmacokinetic properties. Working with collaborators, we are investigating the possible mechanism(s) of action of these compounds by utilising whole genome DNA microarray analysis.
